You shouldn't have to use /loadui, in fact it can sometimes cause some weird things to go wrong. If you've got the UI installed in it's own directory under UI/, then just go into eq2.ini in the program directory and edit the eq2.ini file to have:

cl_ui_skinname yourdirectory
cl_ui_subdir UI/

Substitute the folder name you used for your UI for yourdirectory. That should make EQ2 load your custom UI on startup. If there's a problem check the ui.log file in the program directory, that's where the error messages will be.
