This would actually happen if the database is opened by another instance. Make sure there is no other thread of javaw.exe running before launching the updater (reboot usually helps).
Maybe it does not work because the updater crashes while accessing the database and so the process stays active even after closing the window. In that case do a reboot and delete the files in "%appdata%\ProfitUI Reborn Updater" manually. Maybe zip them up and send them to me before as it would be interesting what got messed up in them.
